RESUMO
O diabetes mellitus é uma patologia caracterizada por hiperglicemia e distúrbios no
metabolismo de carboidratos, gorduras e proteínas, que estão associados a uma deficiência
absoluta ou relativa na secreção de insulina pelas células beta do pâncreas. Embora seja
uma doença de origem endócrina, suas principais manifestações são de origem metabólica,
podendo haver complicações vasculares e neuropáticas. Esses distúrbios em longo prazo
são responsáveis por complicações crônico-degenerativas tais como: microangiopatias
(retinopatia e nefropatia), macroangiopatias e a neuropatia diabética. As neuropatias
diabéticas manifestam-se em função de lesões de fibras nervosas periféricas por alterações
na microcirculação dos nervos (vasa nervorum), e no metabolismo dos neurônios,
resultando em sérias repercussões às áreas inervadas. Em nível gastrintestinal leva à
modificações motoras, sensoriais e na função reflexa desse sistema, podendo ocasionar
gastroparesia, diarréia, constipação, megacolon, lentidão do trânsito gastrointestinal, estase
e dilatação gástrica com diminuição ou aumento de contrações peristálticas. Vários
mecanismos são descritos para tentar explicar esta patologia, inclusive o estresse oxidativo.
Este mecanismo, quando associado ao diabetes mellitus, é aumentado e pode ter um
importante papel na patogenia das complicações do diabetes. O aumento do estresse
oxidativo pode ser resultado da alta produção de precursores de espécie reativa de oxigênio
(ROS) e/ou baixa eficiência de sistemas enzimáticos que os combatem. O tecido nervoso
pode ser afetado pelos radicais livres através peroxidação lipídica das membranas do
axônio e das células de Schwann, levando a uma diminuição da função das células
nervosas. A glicosilação autoxidativa, via do poliol, alterações nos níveis de mediadores
inflamatórios e redução dos antioxidantes são também mecanismos potenciais para o
aumento do estresse oxidativo. A ativação da via do poliol devido à hiperglicemia é um
outro fator determinante no desenvolvimento da neuropatia diabética. Esta via resulta no
acúmulo de sorbitol e frutose nos nervos, a qual é acompanhada por decréscimo de
mioinositol e inibição da bomba Na
+
/K
+
ATPase, resultando em edema, aumento de
mielina, disjunção axo-glial e degeneração do nervo. Por outro lado, a enzima aldose
redutase para converter glicose em sorbitol utiliza o NADPH. Com a depleção de NADPH,
as enzimas que dele necessitam podem ser inibidas. Dentre várias, incluímos a glutationa
redutase que necessita de NADPH para regenerar a glutationa oxidada em reduzida. Deste
modo, a diminuição nos níveis de glutationa reduzida aumenta a susceptibilidade das
células endoteliais ao dano causado pelo peróxido de hidrogênio. Estudos têm demonstrado
que o diabetes mellitus pode reduzir o número de neurônios mioentéricos. Drogas que
reduzem o estresse oxidativo podem ter um papel relevante no tratamento das complicações
neurológicas do diabetes mellitus. A glutamina é um aminoácido não essencial, sintetizado
a partir das necessidades corporais, sendo a forma mais abundante encontrada no corpo.
Um fator de particular interesse na suplementação com glutamina no diabetes mellitus é
que esse aminoácido é de grande importância em estados catabólicos e também precursora
da glutationa. O objetivo deste estudo foi avaliar o efeito da suplementação com l-
glutamina sobre os neurônios mioentéricos imunomarcados pela miosina-V e da túnica
mucosa do íleo de ratos diabéticos. Foram utilizados cinco grupos de animais (Rattus
norvegicus): normoglicêmico (UN), normoglicêmico tratado com l-glutamina (NG),
diabéticos (UD), diabéticos tratados com l-glutamina após o 4

dia da indução do diabetes
(DG4) e diabéticos tratados com l-glutamina após o 45
o
dia da indução do diabetes (DG45).
Para a indução do DM foi administrada a droga estreptozootocina (35 mg/Kg), sendo
mantida esta condição durante 120 dias. Aos 210 dias de idade, os animais foram
anestesiados, mortos e amostras do íleo foram coletadas. Para análise da população
